ABSTRACT

The present work evaluated the immunomodulatory effect of thalidomide (Thal) at different doses on tumor-associated macrophages (TAMs) using a mouse model of human breast cancer. Mice were inoculated with 4T1 cells in the left flank and treated with Thal once a day at concentrations of 50, 100, and 150mg/kg body weight from the 5th day until the 28th day of tumor inoculation. The tumors were sized, proliferation index and TAMs count were evaluated in primary tumors and metastatic lungs. In addition, the metastasis rate was evaluated in the lungs. Thal at 150mg/kg significantly decreased tumor growth, proliferation index, and TAMs infiltration in primary tumors. Conversely, a higher number of TAMs and lower proliferation index were observed in metastatic lungs in mice treated with 150mg/kg of Thal. Furthermore, Thal at 150mg/kg significantly decreased the metastatic nodules in the lungs. Our findings demonstrated that Thal treatment considerably decreased the primary tumor and lung metastasis in mice associated with different TAM infiltration effects in these sites.(AU)


No presente trabalho, foi avaliado o efeito imunomodulador de diferentes doses de talidomida em macrófagos associados ao tumor (TAMs), em um modelo murino de câncer de mama. Camundongos foram inoculados com células 4T1, na região do flanco esquerdo, e tratados com talidomida, uma vez ao dia, nas doses de 50, 100 e 150mg/k, por massa corporal, do quinto dia ao 28º dia de inoculação tumoral. Os tumores foram medidos, o índice de proliferação celular e a contagem de TAMs foram avaliados nos tumores primários e nos pulmões com metástases. Além disso, a taxa de metástases pulmonares também foi avaliada. A talidomida na dose de 150mg/kg diminuiu significativamente o crescimento tumoral, o índice de proliferação celular e a infiltração de TAMs nos tumores primários. Por outro lado, maior número de TAMs e menor índice de proliferação celular foram observados nos pulmões metastáticos, em camundongos tratados com 150mg/kg de talidomida. Ademais, a talidomida na dose de 150mg/kg diminuiu significativamente os nódulos metastáticos nos pulmões. Os resultados demonstraram que o tratamento com talidomida diminuiu o crescimento tumoral e as metástases pulmonares em camundongos, associado com diferentes efeitos na infiltração de TAMs nesses locais.(AU)

ABSTRACT

Lymph node metastases are an independent prognosis factor in gastric carcinoma (GC) patients. Radical lymphadenectomy can improve survival but it can also increase surgical morbidity. As a principle, sentinel node (SN) navigation surgery can avoid unnecessary lymphadenectomy without compromising prognosis. In this pilot study, 24 patients with untreated GC were initially screened for SN navigation surgery, of which 12 were eligible. Five patients had T2 tumors, 5 had T3 tumors and 2 had T1 tumors. In 33% of cases, tumor diameter was greater than 5.0 cm. Three hundred and eighty-seven lymph nodes were excised with a median of 32.3 per patient. The SN navigation surgery was feasible in all patients, with a median of 4.5 SNs per patient. The detection success rate was 100%. All the SNs were located in N1 and N2 nodal level. In 70.9% of cases, the SNs were located at lymphatic chains 6 and 7. The SN sensitivity for nodal staging was 91.6%, with 8.3% of false negative. In 4 patients who were initially staged as N0, the SNs were submitted to multisection analyses and immunohistochemistry, confirming the N0 stage, without micrometastases. In one case initially staged as negative for nodal metastases based on SN analyses, metastases in lymph nodes other than SN were found, resulting in a 20% skip metastases incidence. This surgery is a reproducible procedure with 100% detection rate of SN. Tumor size, GC location and obesity were factors that imposed some limitations regarding SN identification. Results from nodal multisection histology and immunohistochemistry analysis did not change initial nodal staging.

ABSTRACT

The present study analyzed the efficiency of the photo-electro-oxidation process as a method for degradation and inactivation of adenovirus in water. The experimental design employed a solution prepared from sterile water containing 5.107 genomic copies/L (gc/L) of a standard strain of human adenovirus type 5 (HAdV-5) divided into two equal parts, one to serve as control and one treated by photo-electro-oxidation (PEO) for 3 hours and with a 5A current. Samples collected throughout the exposure process were analyzed by real-time polymerase chain reaction (qPCR) for viral genome identification and quantitation. Prior to gene extraction, a parallel DNAse treatment step was carried out to assess the integrity of viral particles. Integrated cell culture (ICC) analyses assessed the viability of infection in a cell culture. The tested process proved effective for viral degradation, with a 7 log10 reduction in viral load after 60 minutes of treatment. The DNAse-treated samples exhibited complete reduction of viral load after a 75 minute exposure to the process, and ICC analyses showed completely non-viable viral particles at 30 minutes of treatment.


Resumo O presente estudo analisou a eficiência do processo de fotoeletrooxidação como metodologia para a degradação e inativação de adenovírus em água. A concepção experimental emprega uma solução preparada a partir de água estéril contendo 5,107 cópias genômicas/L (gc/L) de uma amostra padrão de adenovírus humano tipo 5 (HAdV-5), dividida em duas partes iguais, uma para servir como controle e outra tratada por fotoeletrooxidação (PEO) durante 3 horas e com uma corrente de 5A. As amostras recolhidas durante o processo de exposição foram analisadas por PCR quantitativo em tempo real (qPCR) para identificação e quantificação do genoma viral. Antes da extração de ácidos nucleicos, um passo de tratamento com DNAse paralelo foi realizado para avaliar a integridade das partículas virais. Um ensaio de qPCR integrado à cultura de células (ICC-qPCR) permitiu analisar a viabilidade de infecção em uma cultura de células. O processo mostrou-se eficaz testada para a degradação viral, com uma redução de 7 log10 da carga viral após 60 minutos de tratamento. As amostras tratadas com DNAse exibiram redução completa da carga viral após uma exposição de 75 minutos ao processo, e a análise de ICC-qPCR mostrou partículas virais completamente não-viáveis ​​em 30 minutos de tratamento.

ABSTRACT

The advanced oxidation process (AOP) is used to increase the treatment efficiency of effluents however, it is necessary to compare the toxicity of treated and untreated effluents to evaluate if the decontamination process does not cause any biological harm. Cultured cells have been previously used to assess the genotoxic and cytotoxic potential of various compounds. Hence, the aim of this work was to assess the applicability of cytotoxicity assays to evaluate the toxicity related to the AOP treatment. Samples of an industrial effluent were collected after their treatment by a conventional method. Cytotoxicity of standard and AOP treated effluents was assessed in CRIB and HEp-2 cell line using the MTT and neutral red assays. We observed decrease at cell viability in the both assays (50% MTT and 13% NRU) when cells were exposed to the AOP treatment in the highest concentration. Thus, cytotoxic assays in cultured cells can be explored as an useful method to evaluate toxicity as well as to optimize effluents treatment process.


Resumo O processo de oxidação avançada (POA) tem sido usado para aumentar a eficiência do tratamento de efluentes; no entanto, é necessário comparar a toxicidade de efluentes tratados e não tratados para avaliar se o processo de descontaminação não é capaz de causar algum risco biológico. Cultivos celulares têm sido utilizados para avaliar o potencial genotóxico e citotóxico de vários compostos. Assim, o objetivo deste trabalho foi avaliar a aplicabilidade de ensaios de citotoxicidade para avaliar a toxicidade relacionada ao tratamento com POA. As amostras de um efluente industrial foram recolhidas após o tratamento por um método convencional. A citotoxicidade dos efluentes padrão e tratado com POA foi avaliada nas linhagens celulares CRIB e HEp-2 usando os ensaios do MTT e do vermelho neutro. Observou-se diminuição da viabilidade celular em ambos os ensaios (50% MTT e 13% VN) quando as células foram expostas à concentração mais elevada do efluente tratado com POA. Assim, os ensaios de citotoxicidade em cultivos celulares podem ser explorados como um método útil para avaliar a toxicidade, bem como para otimizar os processos de tratamento de efluentes.

ABSTRACT

This research aimed to evaluate the air quality, by determining the concentrations of PM2.5-10, PM2.5 and the metallic elements Al, Ba, Cd, Cr, Cu, Fe, Mn, Ni, Pb, Zn and Hg in the leaf part of ryegrass (Lolium multiflorum) in an area close to Schmidt Stream, at the lower section of Sinos River Basin (SRB), in a research campaign of six months, from October 2013 to March 2014. The particles collected in the PM sampling were analyzed by Scanning Electron Microscopy (SEM) combined with Energy Dispersive X-ray Spectrometry (EDS), in order to study their morphology and chemical composition. The mean concentration of PM2.5-10 was 9.1 µg m–3, with a range of 2.2 µg m–3 to 15.4 µg m–3 and the mean concentration of PM2.5was 4.7 µg m–3, with a range of 1.9 µg m–3 to 8.2 µg m–3. Concentrations of metallic elements, especially Pb, Cr and Zn, were classified as Class 4 (very high pollution levels), according to the classification proposed by Klumpp et al. (2004). Chemical and morphological analysis of PM revealed the presence of particles of biological origin, soot (Cr, Fe, Ni, Zn, Cd, Hg and Pb), salts (KCl) and soil resuspension (Al and Si). The integrated study methodology, employing environmental variables, such as PM and ryegrass, can be of help in the preparation of wide-ranging environmental diagnoses, in addition providing information needed to develop precautionary measures designed to minimize the effects of atmospheric pollution that takes into consideration the environment’s supportive capacity and environmental quality.


Resumo O objetivo desta pesquisa foi avaliar a qualidade do ar, por meio da determinação das concentrações do MP2,5-10, MP2,5 e dos elementos metálicos Al, Ba, Cd, Cr, Cu, Fe, Mn, Ni, Pb, Zn e Hg na folha do azevém (Lolium multiflorum) em uma área próxima ao Arroio Schmidt, no trecho inferior da Bacia do Rio dos Sinos (BRS), em uma campanha de seis meses de pesquisa, de outubro de 2013 a março de 2014. As partículas coletadas na amostragem do MP foram analisadas por Microscopia Eletrônica de Varredura (MEV), combinada com Espectrometria de Energia Dispersiva de Raios X (EDS), a fim de estudar sua morfologia e composição química. As concentrações médias de MP2,5-10 e MP2,5 foram de 9,1 µg m–3 e 4,7 µg m–3, respectivamente. As concentrações médias dos elementos metálicos Pb, Cr e Zn na parte foliar do azevém foram de 13,58 mg kg–1, 5,26 mg kg–1 e 88,80 mg kg–1, respectivamente, caracterizando a área como Classe 4 (nível muito elevado de poluição), conforme classificação proposta por Klumpp et al. (2004). A caracterização química e morfológica das partículas coletadas revelou a presença de fuligem (Cr, Fe, Ni, Zn, Cd, Hg e Pb), materiais biológicos, cristais salinos (KCl) e partículas ressuspensas de poeira do solo (Al e Si). Estudos integrados empregando variáveis ambientais como o MP e o biomonitoramento com azevém podem auxiliar na elaboração de diagnósticos ambientais robustos, além de fornecer informações para o desenvolvimento de medidas de precaução que considerem a capacidade de suporte do meio ambiente e que visem à minimização dos efeitos prejudiciais da poluição atmosférica.

ABSTRACT

Worldwide environmental pollution is increasing at the same rate as social and economic development. This growth, however, is disorganized and leads to increased degradation of water resources. Water, which was once considered inexhaustible, has become the focus of environmental concerns because it is essential for life and for many production processes. This article describes monitoring of the water quality at three points along the Sinos River (RS, Brazil), one in each of the upper, middle and lower stretches. The points were sampled in 2013 and again in 2014. The water samples were analyzed to determine the following physical and chemical parameters plus genotoxicity to fish: metals (Cr, Fe, Al), chemical oxygen demand, biochemical oxygen demand, chlorides, conductivity, total suspended solids, total phosphorous, total and fecal coliforms, pH, dissolved oxygen, turbidity, total Kjeldahl nitrogen nitrate and ammoniacal nitrogen. Genotoxicity was tested by exposing individuals of the species Astyanax jacuhiensis to water samples and then comparing them with a control group exposed to water from the public water supply. The results confirmed the presence of substances with genotoxic potential at the sample points located in the middle and lower stretches of the river. The results for samples from the upper stretch, at P1, did not exhibit differences in relation to the control group. The physical and chemical analyses did not detect reductions in water quality in the lower stretch, as had been expected in view of the large volumes of domestic and industrial effluents discharged into this part of the river.


Resumo A poluição ambiental aumenta mundialmente, no mesmo ritmo que o desenvolvimento social e econômico. Este crescimento é, muitas vezes, desorganizado e proporciona um aumento da degradação dos recursos hídricos. Água, que já foi considerado inesgotável, tornou-se o foco das preocupações ambientais, pois é essencial para a vida e para muitos processos de produção. Este artigo aborda o monitoramento da qualidade da água, em três pontos do Rio dos Sinos, compreendendo o trecho superior, médio e inferior. Os pontos foram monitorados entre 2013 e 2014. As amostras de água foram analisadas para determinar os parâmetros físico-químicos complementarmente a genotoxicidade em peixes: metais (Cr, Fe, Al), demanda química de oxigenio, demanda bioquímica de oxigênio, cloretos, condutividade, sólidos suspensos totais, fósforo total, coliformes totais e fecais, pH, oxigênio dissolvido, turbidez, nitrogênio total Kjeldahl, nitrato e nitrogênio amoniacal. A avaliação de genotoxicidade foi conduzida pela exposição à água de diferentes pontos do rio de indivíduos da espécie Astyanax jacuhiensis. O grupo controle foi exposto à água de abastecimento público. Os resultados confirmam a presença de substâncias com potencial genotóxico nos pontos localizados nos trechos médio e inferior. No trecho superior, ponto 1, não houve diferenças em relação ao grupo controle. A diminuição da qualidade da água do trecho inferior quando comparada com o superior também foi confirmada através das análises físico-químicas, as quais indicaram uma redução na qualidade da água nos trechos inferiores, como o esperado devido ao grande escoamento de esgotos domésticos e industriais neste trecho do rio.

Worldwide environmental pollution is increasing at the same rate as social and economic development. This growth, however, is disorganized and leads to increased degradation of water resources. Water, which was once considered inexhaustible, has become the focus of environmental concerns because it is essential for life and for many production processes. This article describes monitoring of the water quality at three points along the Sinos River (RS, Brazil), one in each of the upper, middle and lower stretches. The points were sampled in 2013 and again in 2014. The water samples were analyzed to determine the following physical and chemical parameters plus genotoxicity to fish: metals (Cr, Fe, Al), chemical oxygen demand, biochemical oxygen demand, chlorides, conductivity, total suspended solids, total phosphorous, total and fecal coliforms, pH, dissolved oxygen, turbidity, total Kjeldahl nitrogen nitrate and ammoniacal nitrogen. Genotoxicity was tested by exposing individuals of the species Astyanax jacuhiensis to water samples and then comparing them with a control group exposed to water from the public water supply. The results confirmed the presence of substances with genotoxic potential at the sample points located in the middle and lower stretches of the river. The results for samples from the upper stretch, at P1, did not exhibit differences in relation to the control group. The physical and chemical analyses did not detect reductions in water quality in the lower stretch, as had been expected in view of the large volumes of domestic and industrial effluents discharged into this part of the river.


Resumo A poluição ambiental aumenta mundialmente, no mesmo ritmo que o desenvolvimento social e econômico. Este crescimento é, muitas vezes, desorganizado e proporciona um aumento da degradação dos recursos hídricos. Água, que já foi considerado inesgotável, tornou-se o foco das preocupações ambientais, pois é essencial para a vida e para muitos processos de produção. Este artigo aborda o monitoramento da qualidade da água, em três pontos do Rio dos Sinos, compreendendo o trecho superior, médio e inferior. Os pontos foram monitorados entre 2013 e 2014. As amostras de água foram analisadas para determinar os parâmetros físico-químicos complementarmente a genotoxicidade em peixes: metais (Cr, Fe, Al), demanda química de oxigenio, demanda bioquímica de oxigênio, cloretos, condutividade, sólidos suspensos totais, fósforo total, coliformes totais e fecais, pH, oxigênio dissolvido, turbidez, nitrogênio total Kjeldahl, nitrato e nitrogênio amoniacal. A avaliação de genotoxicidade foi conduzida pela exposição à água de diferentes pontos do rio de indivíduos da espécie Astyanax jacuhiensis. O grupo controle foi exposto à água de abastecimento público. Os resultados confirmam a presença de substâncias com potencial genotóxico nos pontos localizados nos trechos médio e inferior. No trecho superior, ponto 1, não houve diferenças em relação ao grupo controle. A diminuição da qualidade da água do trecho inferior quando comparada com o superior também foi confirmada através das análises físico-químicas, as quais indicaram uma redução na qualidade da água nos trechos inferiores, como o esperado devido ao grande escoamento de esgotos domésticos e industriais neste trecho do rio.

ABSTRACT

The Sinos River Basin (SRB) is located in the northeastern region of the state of Rio Grande do Sul (29º20' to 30º10'S and 50º15' to 51º20'W), southern Brazil, and covers two geomorphologic provinces: the southern plateau and the central depression. It is part of the Guaíba basin, has an area of approximately 800 km 2 and contains 32 counties. The basin provides drinking water for 1.6 million inhabitants in one of the most important industrial centres in Brazil. This study describes different water quality indices (WQI) used for the sub-basins of three important streams in the SRB: Pampa, Estância Velha/Portão and Schmidt streams. Physical, chemical and microbiological parameters assessed bimonthly using samples collected at each stream source were used to calculate the Horton Index (HI), the Dinius Index (DI) and the water quality index adopted by the US National Sanitation Foundation (NSF WQI) in the additive and multiplicative forms. These indices describe mean water quality levels at the streams sources. The results obtained for these 3 indexes showed a worrying scenario in which water quality has already been negatively affected at the sites where three important sub-basins in the Sinos River Basin begin to form.


A Bacia Hidrográfica do Rio dos Sinos (BHRS) está localizada na região Nordeste do Estado do Rio Grande do Sul (29º20' to 30º10'S and 50º15' to 51º20'W), na região Sul do Brasil, envolvendo duas regiões geomorfológicas, o planalto Sul e a depressão central. Esta bacia faz parte da Bacia do Guaíba e tem uma área de aproximadamente 800 km 2, atingindo 32 municípios. A BHRS fornece água de abastecimento para cerca de 1.6 milhões de habitantes e para um dos mais importantes centros industriais do Brasil. Este artigo apresenta a determinação de diferentes índices de qualidade de água (IQA) para as regiões de nascente de três importantes sub-bacias pertencentes à BHRS: arroios Pampa, Estância Velha/Portão e Schmidt. Utilizando parâmetros físicos, químicos e microbiológicos, analisados em amostras coletadas bimestralmente em cada ponto próximo à nascente, foram calculados os índices de Horton (IH), de Dinius (ID) e o índice de qualidade de água da National Sanitation Foundation, EUA (IQANSF) usando os métodos somatório e produtório. Os resultados foram analisados para verificação da qualidade da água nos pontos amostrais a fim de comparar e classificar a água na região das nascentes das três sub-bacias. Uma avaliação conjunta dos índices calculados indicou que a água nos três pontos amostrais apresenta qualidade média. Os resultados dos três índices convergem a um cenário preocupante que indica que, desde os pontos iniciais de formação de três importantes sub-bacias do rio dos Sinos, já ocorrem problemas de qualidade de água.

ABSTRACT

Na atualidade, o sorovar Copenhageni é o representante do sorogrupo Icterohaemorrhagiae, mantido por roedores sinantrópicos, que tem prevalecido nos cães e seres humanos das grandes metrópoles brasileiras. A despeito de alguns autores sugerirem a existência de proteção cruzada entre sorovares incluídos em um mesmo sorogrupo esta condição ainda não foi suficientemente esclarecida para os sorovares Icterohaemorrhagiae e Copenhageni. No presente trabalho cães adultos com dois a seis anos de idade primo-vacinados com três doses intervaladas de 30 dias a partir dos 60 dias de idade e revacinados anualmente com vacina anti-leptospirose polivalente contendo os sorovares Canicola, Icterohaemorrhagiae, Grippotyphosa e Pomona foram revacinados com a mesma vacina e aos 30 dias da revacinação foram submetidos aos testes de soroaglutinação microscópica (SAM) e de inibição do crescimento de leptospiras in vitro (TICL), para avaliação comparativa dos níveis de anticorpos produzidos para os sorovares Canicola, Icterohaemorrhagiae e Copenhageni. Os resultados obtidos indicaram que a imunidade conferida pela vacina para o sorovar Icterohaemorrhagiae é mais duradoura que a observada para o sorovar Canicola, já que títulos de anticorpos neutralizantes >1,0 log10 foram observados antes do reforço vacinal não havendo substancial aumento após a revacinação. Quanto ao sorovar Canicola, a revacinação resultou em considerável aumento do título de anticorpos neutralizantes quando comparado ao momento anterior a revacinação (p=0,001). A análise dos valores encontrados após a revacinação demonstrou claramente que cães revacinados com bacterina produzida com o sorovar Icterohaermorrhagiae não apresentam aumento do título de anticorpos inibidores do crescimento contra o sorovar Copenhageni, em nível suficiente para inibir o crescimento de leptospiras. Apesar disso, os títulos de anticorpos inibidores de crescimento anti-Copenhageni encontrados antes e após a revacinação demonstraram que, pelo menos certo grau de proteção contra a infecção por esse sorovar pode ser esperado para os cães vacinados com bacterinas do sorovar Icterohaemorrhagiae, não sendo, no entanto, uma proteção cruzada completa.


Currently, the serovar Copenhageni is the representative of serogroup Icterohaemorrhagiae maintained in synanthropic rodents found most frequently in dogs and humans in metropolitan areas of Brazil. Despite some authors have suggested the existence of cross-protection between serovars included in the same serogroup, this condition has not yet been sufficiently clarified for serovars Icterohaemorrhagiae and Copenhageni. In the present work, 2 to 6-year-old dogs, vaccinated at 60, 90 and 120 days of age and thereafter, revaccinated annually with commercial vaccine containing Canicola, Icterohaemorrhagiae, Grippotyphosa and Pomona bacterins were evaluated as to the immune status against leptospirosis before and 30 days after revaccination. Mycroscopic agglutination test (MAT) and in vitro growth inhibition test (GIT) were performed to search for agglutinating anti-Leptospira antibodies and neutralizing anti-Leptospira antibodies, respectively for serovars Canicola and Icterohaemorrhagiae, and additionally, for serovar Copenhageni, not included in the vaccine. The results showed that the immunity conferred by the vaccine to serovar Icterohaemorrhagiae is more lasting than that observed for serovar Canicola, since neutralizing antibody titers >1.0 log10 were observed before the booster vaccination with no substantial increase after revaccination. As for the serovar Canicola, revaccination resulted in a considerable increase in neutralizing antibody titer when compared to the one observed previously to the revaccination (p=0.001). The analysis of the data obtained by GIT allowed us to conclude that dogs given vaccine containing Icterohaemorrhagiae bacterin did not produce neutralizing antibodies against serovar Copenhageni enough to inhibit leptopiral growth at the same level as occurred for the homologous serovar. Despite this, the GIT titer found for serovar Copenhageni before and after revaccination showed that at least, some level of protection could be expected for dogs vaccinated with serovar Icterohaemorrhagiae bacterin, not a complete cross protection.

ABSTRACT

"Quina" (Strychnos pseudoquina A. St. Hil) is a medicinal plant species from the Brazilian Cerrado. As its seeds show dormancy, they were subjected to the treatments pre-cooling at 5ºC during 7 days, pre-heating at 40ºC during 7 days, pre-soaking in sulfuric acid PA during 5 and 15 min, pre-soaking in boiling water during 5 and 15 min, pre-soaking in 100 and 200 ppm gibberellic acid during 48 h, pre-soaking in distilled water during 24 and 48 h, and mechanical scarification to break dormancy. Counts were daily conducted from the 2nd day after the experiment implementation until the germination stabilization at the 65th day. The germination speed index (GSI) and the germination percentage were evaluated. Germination rates above 96 percent were reached in seeds pre-soaked in water during 48 h and substrate moistened with water or KNO3.


A quina (Strychnos pseudoquina A. St.-Hil) é espécie medicinal do Cerrado Brasileiro. As sementes apresentam dormência e, por isso, foram submetidas aos tratamentos pré-resfriamento a 5ºC durante 7 dias, pré-aquecimento a 40ºC durante 7 dias, pré-embebição em ácido sulfúrico P.A. durante 5 e 15 minutos, pré-embebição em água fervente durante 5 e 15 minutos, pré-embebição em ácido giberélico a 100 e 200 ppm, durante 48 horas, pré-embebição em água destilada durante 24 e 48 horas e escarificação mecânica, visando à superação da dormência. Foram realizadas contagens diárias entre o 2º dia após a implantação do experimento até a estabilização da germinação, que ocorreu no 65º dia. Foram avaliados o índice de velocidade de germinação (IVG) e a porcentagem de germinação. Taxas de germinação acima de 96 por cento foram atingidas em sementes pré-embebidas em água durante 48 horas e substrato umedecido com água ou KNO3.

ABSTRACT

Calcium (Ca2+) is a versatile second messenger that regulates a wide range of cellular functions. Although it is not established how a single second messenger coordinates diverse effects within a cell, there is increasing evidence that the spatial patterns of Ca2+ signals may determine their specificity. Ca2+ signaling patterns can vary in different regions of the cell and Ca2+ signals in nuclear and cytoplasmic compartments have been reported to occur independently. No general paradigm has been established yet to explain whether, how, or when Ca2+ signals are initiated within the nucleus or their function. Here we highlight that receptor tyrosine kinases rapidly translocate to the nucleus. Ca2+ signals that are induced by growth factors result from phosphatidylinositol 4,5-bisphosphate hydrolysis and inositol 1,4,5-trisphosphate formation within the nucleus rather than within the cytoplasm. This novel signaling mechanism may be responsible for growth factor effects on cell proliferation.

ABSTRACT

A Proteína Catiônica Eosinofílica (ECP) foi purificada de células humanas, pela primeira vez, em 1971 e, após cinco anos, foi identificada como sendo uma proteína granular eosinofílica que possui uma potente toxicidade para uma variedade de helmintos, bactérias e outros microrganismos. O objetivo deste trabalho foi diagnosticar as infecções por enteroparasitas correlacionando com níveis séricos de proteína catiônica eosinofílica e eosinofilia. Foram estudadas 150 crianças de ambos os sexos, com idade variando entre 3 e 6 anos, residentes no mesmo bairro e frequentadoras de creche. A Infecção Parasitária foi investigada por Exames de fezes pelosMétodos de Hoffmann, Pons & Janer e o de Baermann-Morais. Os níveis séricos de proteína catiônica eosinofílica foram determinadospor Fluoroenzimaimunoensaio, utilizando o Kit Unicap (Pharmacia & Upjonh) e a contagem de eosinófilos realizada em esfregaço sanguíneo corado pelo Leishman. Das crianças estudadas 140 (93,3%) apresentaram infecção por enteroparasitas e 10 (6,7%) apresentaram ausência de ovos e larvasde helmintos e cistos de protozoários. Cento e quarenta e oito amostras de soro foram analisadas para determinar os níveis de proteína catiônica eosinofílica e os resultados obtidos mostraram que foi 45,45μg/L a mediana das concentrações observadas e que com relação aos níveis de ECP em crianças parasitadas e não parasitadas por helmintos observou-se que as crianças parasitadas apresentaramconcentrações (MD=52,20μg/L) significantemente mais elevadas do que as não parasitadas (MD=29,70μg/L) e que houve uma correlação positiva entre níveis séricos de ECP e eosinófilos ( p< 0,0001 e r= 0,57).


The eosinophil cationic protein was purified from human cells, for the fist time in 1971, after five years, it was identified as being a eosinophil granular protein that is potencially toxic for helminthics, bacteria and other microorganisms. The purpose of this study was to diagnose the enteroparasitics infections co-related to the serum levels of eosinophil cationic protein and eosinophilia. One hundred and fifty children form both sexes, ages varying from 3 to 6 years old who live in the same neighbourhood and go to the same nursery were observed. The parasitic infection was investigated by faecal examinations using the Hoffmann, Pons & Janer and Baermann-morais methods. The serum cationic protein levels were determined by fluorimmunassay using the unicap kit (Pharmacia & Upjohn) and the counting of eosinophis was made by Leishman- stained smears. From the studied children 140 (93,3%) were infected by enteroparasitics and 10 (6,7%) showed no signs of eggs, helminthics, larva,and protozoarios cysts. One hundred and light samples of serum were analysed to determine the levels of eosinophil cationis protein and the obtaind results have showed that 45,45 ìg/L was the overage of the studied concentrations and that the relationship between the levels of eosinophil cationic protein (ECP) in parasited and non-parasited children was that the parasited children showed significantly hight concentrations (MD=52,20ìg/L) than the non-parasited children (MD=29,70 ìg/L). The results also have showed that there was a positive correlation between the serum levels of ECP and eosinophis ( p<0,0001 and r = 0,57).

Aberrant crypt foci (ACF) in the colon of carcinogen-treated rodents are considered to be the earliest hallmark of colon carcinogenesis. In the present study the relationship between a short-term (4 weeks) and medium-term (30 weeks) assay was assessed in a model of colon carcinogenesis induced by dimethylhydrazine (DMH) in the rat. Six-week-old male Wistar rats were given subcutaneous injections of DMH (40 mg/kg) twice a week for 2 weeks and killed at the end of the 4th or 30th week. ACF were scored for number, distribution pattern along the colon and crypt multiplicity in 0.1 percent methylene-blue whole-mount preparations. ACF were distinguished from normal crypts by their larger size and elliptical shape. The incidence, distribution and morphology of colon tumors were recorded. The majority of ACF were present in the middle and distal colon of DMH-treated rats and their number increased with time. By the 4th week, 91.5 percent ACF were composed of one or two crypts and 8.5 percent had three or more crypts, while by the 30th week 46.9 percent ACF had three or more crypts. Thus, a progression of ACF consisting of multiple crypts was observed from the 4th to the 30th week. Nine well-differentiated adenocarcinomas were found in 10 rats by the 30th week. Seven tumors were located in the distal colon and two in the middle colon. No tumor was found in the proximal colon. The present data indicate that induction of ACF by DMH in the short-term (4 weeks) assay was correlated with development of well-differentiated adenocarcinomas in the medium-term (30 weeks) assay

ABSTRACT

The objective of the present investigation was to determine the course of maternal blood glucose levels in pregnant rats and its repercussions on the glucose levels an pancreas of their newborn pups. Diabetes was induced by alloxan (42mg/Kg body weight) and steptozotocin (40mg/Kg). Sixty-two pregnant Wistar rats weighing 180 to 250 g were divided into a control group and two groups with moderate (120 to 200 mg/dl glucose) and severe diabetes (greater than 200 mg/dl glucose), respectively. Blood glucose levels were measured in the dams on the 1st, 14th, and 21st days of pregnancy and in the pups at birth. The results were pooled for each litter. The fetal pancrases were removed after cesarian section performed on the 21st day of pregnancy, pooled for each litter and processed for histopathologic examination by light microscopy. Maternal blood glucose levels were significantly increased compared with the first day of pregnancy in both normal and diabetic ratsd starting on the 14 th day of pregnancy. Fetal blood glucose levels correlated with maternal levels. The histopathologic changes characterized by vacuolization and basophilia of the cytoplasm of endocrine pancreas of newborn pups from dams with moderate or severe diabetes suggested pancreatic hyperactivity

ABSTRACT

1. The interaction between experimental protein deprivation and natural intestinal infection by Giardia lamblia was studied in terms of its effects on the intraepithelial lymphocyte (IEL) population and morphology of the jejunal mucosa of rats of different ages. 2. Young, adult and old male Wistar rats received a protein-deficient diet (2 casein) or a control diet (20 casein) for 42 days. Mucosal height and the number of lymphocytes located among 500 consecutive epithelial cells (EC) along the villi or crossing the basement membrane were determined in PAS-stained jejunal fragments. 3. The number of IEL increased progressively with animal age, from 14 to 25 per 100 epithelial cells, with significant differences between age ranges. However, the number of IEL did not differ between control and protein-deficient rats in any of the age groups. The proportion of lymphocytes crossing the basement membrane was approximately two-fold greater in young (2.8/100 EC) and adult (5.8/100 EC) protein-deficient animals than in their respective controls (1.6 and 2.8/100 EC). The intensity of parasite colonization was moderate, from 3 to 5/100 EC and did not differ between groups. The pattern of morphologic changes of jejunal mucosa in protozoal infection did not differ between control and protein-deficient animals in any of the three age groups. 4. We conclude that intestinal infection with Giardia lamblia probably stimulated the local immune response, masking the reduction of the IEL population induced by protein deficiency. The increase in lymphocyte numbers with age may be related to prolonged antigenic stimulation promoted by infection.
